Duke's 2021 NCAA Regional appearance marks the first time in program history that Duke will compete at the event in three consecutive seasons, having also reached regional play in 2018 and 2019. The 2020 season was cut short due to the COVID-19 pandemic and no national tournament was played.

Head coach
Chris Pollard will lead the Blue Devils to a regional for the fourth time in eight seasons (2016, 2018, 2019 and 2021). Prior to Pollard arriving at Duke, the Blue Devils had competed in just five NCAA Tournaments (1952, 1953, 1956, 1957 and 1961).

Duke claimed the ACC's automatic bid into the tournament after winning its first ACC title in 60 years following a 1-0 victory Sunday over third-seeded NC State.
